This two-roomed museum was a bus ride or short car journey from Colonia's old town up the road towards Real de San Carlos. It's not far to travel but as the museums are closed on alternate days, and infrequently due to staffing issues, you might want to call ahead - ask one of the other museums or the hotel to call for you. We arrived and the guide there was a wealth of information about the megafauna that used to live in South America. The fossil evidence is amazing, and shows giant sloths and glyptodonts (large, VW Beetle-sized armadillos) that all lived for millions of years in the region, until the end of the Pleistocene epoch when the ice melted and humans arrived on the scene. There are huge legs, skulls, and 4 different armor cases for the glyptodonts to see. This is a tiny museum, but ask questions and you will be rewarded. They also have fish, and other extinct mammals molars, etc, such as the mammoth. It is on it's way to the bull ring and the railway museum, ad opposite the pirate theme park so there is other stuff to do. The museum is part of the museum experience and so is included in the all-in-one ticket you can purchase in the museums in the old city.
