For 5 euros, you get access to the cathedral, the museum, and the exhibition rooms at Palazzo DePiro. If you want to see the cathedral, you have to buy this ticket.
The museum itself is okay. It has an interesting collection that covers two floors in a lovely old building. My frustration with it was that some of the displays had no explanation as to the contents of the case. However, the picture gallery has some interesting works and there is an extensive coin collection. Coins aren't my thing, but that room seems to be well laid out with a fair amount of detail about the displays, so enthusiasts will likely enjoy it.
I'm not sure this is a "must see" when in Mdina, but those pressed for time could target whatever collection(s) within the museum that interests them. If you want to see the whole museum, you should be able to comfortably do so in an hour.
Not fully accessible to the mobility impaired, but if you can handle a flight of stairs, you should be okay. Moving about within the museum is easy.
