Toronto to Buffalo

I need to arrive to Buffalo from Toronto (and from there rent a car).

Do you have an idea regarding the possibility of doing so?

Would it be better to go to Buffalo airport or the city?

For the most convenient location, the airport would be best. All the car rental agencies are located across Genesee St. adjcent to the Buffalo Niagara International Airport.

If I understand your post, you would be renting a car in Buffalo. It is usually more expensive to rent a car at the airport because of airport taxes added on. I don't know what they would be in Buffalo, but in some cities it makes quite a difference. If you are flying into Buffalo, then yes, get your car at the airport. I wasn't clear from your post how you are arriving in Buffalo and where exactly you will be. If you are renting outside the airport, keep in mind that they will not be open 24/7. So, you need to look at what day and what time you are arriving, as well as where you will arrive in the city.
