tunxi to Yichang

Three ladies will be traveling from tunxi to Yichang to meet cruise up the Yangtze. Should we take bus to Hefei, stay the night and then train to Wuhan to Yichang or go to Wuhan from tunxi and then on to Yichang. How much would a driver cost to Hefei from Tunxi. We will be theer March 15-17

It will be easiest on you if you minimize transfers. A suspect a Tunxi-based driver will charge upwards of RMB 1000 to do the transfer (remember he'll come back empty), but if you have the cash and need the flexibility....maybe worthwhile if there are 3 of you to split the cost. I'd consider taking the public bus Tunxi - Hefei (a little over 4 hours, RMB 114). Then taxi over to Hefei train station and take a fast D train to Yichang (Yichang East station, under 5 hours, RMB 192 2nd class or RMB 231 1st class). There is also an overnight standard train leaving Hefei at 19:21, arriving Yichang 07:11. Soft sleeper on that is RMB 300-ish and saves a hotel night.

You'll need to run the timetable backwards based on the time you need to be in Yichang. If you take a morning bus from Tunxi, then the D3081 train Hefei - Yichang 16:24-21:10 will get you to Yichang the same evening, likely you'd spend a hotel night in Yichang assuming you will board the cruise the next morning. I'd personally choose this method over the overnight train K1513 that gets you to Yichang 07:11 next morning.

If your timing requires you to take an afternoon bus from Tunxi to Hefei, then you should stay the night in Hefei, and take the D3077 leaving the next morning at 08:37, arriving Yichang 13:19. This works if you have a late afternoon or evening sailing time.

Wuhan is a huge city and one big confusing and time-consuming mess. I would try hard to avoid making a transfer there except as a last resort.
