The Airport Express doesn't link to the MTR and has only three stops. The one nearest your hotel is Kowloon Station.

The best ticket to buy is the on-loan Octopus Card. You can use this on buses, trains, ferries etc. When you leave HKG you can all your balance refunded minus a small administration fee.

Bus A21 is a good option to get to Tsim Sha Tsui ($33 and no transfers). You can use the on-loan octopus card to board the bus.

Re #2, the Airport Express does link to the MTR. You can take AE to its first stop at Tsing Yi. At Tsing Yi, change to MTR to Lai King and change again (short cross-platform change) for Tsim Sha Tsui. Airport Express fare to Tsing Yi is HK$60 and the connecting MTR journeys to Tsim Sha Tsui are free with Octopus Card. However, Glacierfed's suggestion of A21 bus is just as good, may take slightly longer but is cheaper and just as convenient.

I suggest you opt for A21. Travel time will be around 1 hour on average.

As mentioned already, you cannot take the MTR directly to TST from the airport. You can, however, take Airport Express to Kowloon Station and transfer to a free shuttle bus to Canarvon Road area. Buses run between around 6am to 11pm. However, this transfer might be just 15 minutes quicker for quite a bit more money.

With Airport Express, it requires one or multiple transfers.

AE to MTR: To Tsing Yi, tranfer, to Lai King, transfer, to Tsim Sha Tsui.

or AE to shuttle bus: one transfer

The speed advantage is not much with the transfer.

I would take the A21 bus, no transfer required, and enjoy the street scenaries along the way.
