Get yourself one of the inexpensive little guide books on Venice, such as the DK "Top Ten Venice", and decide what are things that would interest you. There are hundreds of places of interest to tourists in Venice: only you can decide which ones you should visit.

I would suggest that you begin your first day by taking the #1 vaporetto down the Grand Canal, seeing St. Mark’s Square, visiting St. Mark’s Basilica, touring the Doge’s Palace, and afterwards doing plenty of walking around the backstreets of Venice, well away from the square. See also the Rialto Bridge.

For part of day #2, consider a vaporetto trip to Murano or Burano. (As sydneynick suggested, a guidebook is a good way to plan your own personal itinerary.)

Where you eat depends a lot on your budget.

Since you’ll be staying in Mestre, you might want to have one of your evening meals at Mestre’s historic town square, Piazza Ferretto, which is about a fifteen-minute walk from the Plaza Hotel. You’ll find a number of moderately priced restaurants, cafes, and pastry shops surrounding this attractive piazza.
