We will be arriving from the US but have a connecting flight to Spain. Will we have enough time? We only have one hour. Will we need to go through customs? We are flying Business Class

You go through immigration in Amsterdam, customs in Spain. They are different things.

Yes you go from non-Schengen to Schengen.

The Immigration counters there have a line for short-transfer flights which opens 45m before their scheduled departure, and same line is also open for all business class pax. So you count double ;-) And anyhow if somehow you miss the connection due to delays, KLM will reschedule you on their next flight to same town - generally 2-3 flights per day at least.

KLM will reschedule you IF you are flying with KLM :-)

And you won't have to pick up your luggage until you arrive at your final destination

There is a priority lane for passport control when entering the Schengen area, however there is often a backlup of people trying to get through security where this is no priority lane! I have made it in that time but prefer not to have to.
