Yes, but you have to travel by a slow train. The fast trains do not stop between Rome and Florence.

You take a train from Rome to Orvieto, then another train onwards to Florence. See www.lefrecce.it

Or you could rent a car.

It is very easily done by train and much recommended. Take the train from Florence to Orvieto, there is a funicular, or bus, but the funicular is more fun, from the train station up to the city. Orvieto is a beautiful little city with just enough to see and do to make a perfect 24 hour stopover.

there is no left luggage service at the train station.
