Which is the best town to use as a base for exploring the area and how many days would be necessary?

3 nights and 2 days worked for us, but of course it could have been longer. It depends on how many trip days you have, no? We loved our stay in Manarola...Seaside lodging, mostly surrounded by water and with the very colorful town on one side. It made for some excellent sunset photos and walks. Plus the town had everything we needed for supplies and restaurants, with the train station an easy (and level) walk away.

3 nights depending on your pace, I'd suggest Monterosso or Vernazza

I'm definitely in agreement with the 3 days, but I vote for Monterosso. Beach and easy Intercity train connections, as well as the great hikes from Monterosso-Levanto as well as Monterosso-Vernazza, kayak rentals, and the most "nightlife" get my vote.

Levanto is also nice and well connected by train. And it might be cheaper.
