The buses will of course stick to the autobahns. You'll see a great variety of noise protection walls.
Note that the buses depart from Berlin ZOB (central bus station). Which is however not central. You'll have to calculate 20-30 minutes from central Berlin to get there.
train is also an alternative. Faster and more scenic.
We took the train which was a relaxing and interesting way to see more of Germany. We stopped in Nuremberg on the way thru, staying in the pretty & interesting aldstadt.
The trains are much faster - buses are bound to a speed limit of 100 kph while trains run at up to 250 kph - not dependent on traffic, and more comfortable than any bus. Going by train will take about 6 hours, so you'd have time for a stopover on the way, for example in Erfurt, Bamberg, or Nuremberg. The bus ride will fill most of the day.
