There is a railway station in Bowness-on-Windermere. From Edinburgh, you'd change trains at Oxenholme. However, there aren't many options fro car rental in Bowness. If you want/need a rental car, it might be easier to take the train to Carlisle and rent from there - all the major rental agencies are in Carlisle.

Hire a car in Edinburgh and return it in Manchester - you are going to need a car in the Lakes

What Alan says. If you're going to hire a car might as well just do that in Edinburgh. Carlisle's not really far enough for it to be worth taking a train then hiring a car. Presuming Manchester is after your two night in the lakes?

Yes, agreed. Hiring a car for the whole journey sounds like a far better plan to me too.
