Yes Galle is worth half a day. There is storage for luggage at railway station (at least by renting out a 'retiring room' for a few hours but latest info says the parcel room also offers storage) hence you could use trains as well for this leg.

Are you looking to stay where you are in Mirissa? You're only about 30km to Galle Fort from Mirissa so you can simply catch a taxi to Galle Fort and back, only around a 30 minute trip depending on traffic.
There is also the Train from Mirissa to Galle, although the station is a little in land at Mirissa so you'd probably end up taking a Tuk Tuk to Mirissa Station. Galle Station is right across the road from the Fort, short walk.
For close proximity of Mirissa to Galle Fort, and flexibility of time, take a taxi, 30km possible by Tuk Tuk, could be fun, but probably not my idea of fun.
Other option is simply moving on from Mirissa to Unawatuna, similar beach and setting to Mirissa, as Galle Fort just around the corner.
But for the hassle of moving again, best option is Taxi to/from Galle Fort for at least 1/2 a day as a lot to see there and nice cafes for lunch.
