Fed up?, Uncomfotable?, Tired?
Having sat around the departure lounge for over 2 hours, ants set in well before the flight.

At about half way into the flight.. Realising you still have that many hours to go!!
Same as joe, if in business class in a lie flat, I'm fine throughout. If I am in economy, it's about the six or seven hour mark for me. It's when I start to get very uncomfortable.
Economy, pre flight unless I've got an exit row..
Business/First, not at all.

As long as I have a decent seat in economy, and there's not an "annoying" pax next to me, I can go 13-14 hours without getting antsy. Go prepared for a long flight, don't rely on the IFE to keep you occupied (take an iPAD, load music/podcasts on your phone, take a book) and you should be okay.
