I stay at this hotel regularly, and get there via Southern Cross station from Tullamarine airport. As stated above you can get the Skybus from the Little Bourke Street entrance, but you need to book from the hotel desk about 90 minutes prior. Second option is the short 100 metre walk to Bourke St mall and catch a tram. Trams are free in the city centre. Melbourne's public transport system is second to none, especially in the city centre. If you are feeling like a little light exercise, it is a pleasant 10 to 15 minute walk to Southern Cross Station. Hope that helps
