I stayed at the Ramada for four nights during a conference, with my girlfriend going for the sights during that time. I found the hotels in downtown very expensive, but first was uncertain to book the Ramada due to the descriptions of its surroundings.
Yes, East Colfax is not the best street of Denver, many strange people all day and night. As a woman, you may not wish to walk around alone. As a male or couple, no problem (there are lots of people all day and night, 20 minutes walk to downtown along busy Colfax Avenue). Also, the hotel offers a free shuttle service to any location within 3 miles distance, operating until 10 p.m., and very good for my girlfriend :-).
Our room was large & clean, the bathroom is quite tiny, the hallway is a bit dark, but the hotel has plenty of parking and friendly and helpful staff. If you are sensitive to noise, you may not want a room facing Colfax Avenue, though I found it alright.
All in all, I did not feel unsafe, and the Ramada offers good value for the money, especially in comparison to the downtown hotels.
