Peter Pan / Greyhound run excellent bus service from Boston to NYC and back. The fares are inexpensive and the buses are great. The train also runs, but it's much more expensive. I took Peter Pan in March and was quite pleased. You're advised to avoid the Chinatown buses---Fung Wah in particular has had safety issues, to the point that it's been investigated, etc.
Driving in Boston and NYC is challenging. If you want to avoid it, the bus and public transportation makes it easy. The bus station in Boston is next to South Station, which is right downtown, near the Financial District. IN NYC, the bus terminal is on 42nd Street, right in the thick of things...
The bus trip is about 4.5 hours.
Price matters a lot in terms of where to stay. NYC hotels are very expensive and Boston isn't far behind, but you can get excellent deals.
You might want to look in the recent Boston postings on hotel recommendations or look at the TA hotels lists and come back with some questions about the merits of particular hotels.
