The best way is to rent a car so can visit all around Lake Tahoe

There is a bus directly to South Lake Tahoe casino areas from the Reno Airport:

In Sept it's not that easy to get around the lake. Renting a car will allow you to reach areas of the lake not sewrviced by public transportation.

Highway 395 from the airport to Carson City taking Highway 50 from there to Tahoe (take a left when you get to Tahoe to get to South Shore) is the most direct and easiest route. With a rental car, then you can explore all around Tahoe. The last time I picked someone up at the airport and drove to the casino area at south shore, I believe it took about 1½ hours.

Depends on what you consider "best".

I like the drive over Mt. Rose into Incline Village and down the east shore. It may take 10 or 15 minutes longer, but what views!

From the airport, south on Highway 395, continue up along 431 (Mt. Rose Highway) over the hill to Incline Villlage, take a left to SR 28, continue on south to join Highway 50 into Stateline/South Lake Tahoe

Wow driving to Carson City from the Airport means you miss some of the beautiful scenery - Sand Harbor - on the east side of the lake. I sure wouldn't call that the best route. Incline to South Lake Tahoe has some nice scenery - unlike lots of the west side that has obstructed views of the lake due to buildings. 28 does leave the lake and heads away from the lake for a bit but comes back and offers more nice lake views.

I'd do Mt Rose Highway to 28 in Incline Village. Turn left and continue south on 28 till it meets up with 50 and then into South Lake Taheo.

The route to Incline does not pass through Sand Harbor. That's north of Incline.

The route through Carson City isn't bad- it still has lots of scenic lake views. In the winterish months, I'd never take the Incline route. Far too much like driving in the Arctic for my tastes.

When you come out of the Mt Rose Highway you turn left on 28 and head south. You will pass by Sand Harbor on your way to South Lake Tahoe. The OP is coming in Sept so no worries about winter driving.

I also agree with most of the above posters that the Mt. Rose route is the "best" as you define best. I would also recommend that when you get to Incline Village that you take a very short detour and drive along Lakeshore Blvd. You will see some amazing $$$ homes!!
