Get out of the train station and look around your left shoulder to see a magnificent duomo, to your right the galleria. You can walk straight to the Sforza Castle after the Duomo and Galleria visits. A longer detour to Santa Maria delle Grazie will bring you to see the Last Supper, but not without tickets arranged well in advance.
